Coach Stephen Larkham has rested Slipper and made nine tweaks to the starting side for the first Super Rugby Pacific clash at Ba's Govind Park on Saturday.
In-form fullback Andy Muirhead will captain the side while Test lock Nick Frost will also play for the first time this season and Corey Toole (leg) returns after missing the after-the-siren loss to the Queensland Reds last Saturday.
It means Slipper's record-breaking appearance will likely come at home against the Chiefs next Friday.
The Western Force have also shuffled their deck, making five changes to the starting side and adding code-hopping talent Taj Annan to the bench.
Veteran Wallabies back Kurtley Beale has been promoted from the bench to start at No.12 alongside George Bridge to play the Hurricanes in Napier on Friday.
Scrum half Henry Robertson, winger Bayley Kuenzle, fullback Max Burey and flanker Will Harris are the other changes while No.8 Vaiolini Ekuasi (quad) is out and Nick Champion de Crespigny will slide across to replace him.
Annan, an Australian under-20s representative, joined the Force's wider training squad in pre-season after an NRL stint with Newcastle, having made his Super Rugby debut for the Queensland Reds in 2023.
Tim Ryan has replaced Lachie Anderson (wrist) for the Reds' clash with the Waratahs, while Seru Uru will start at lock for the injured Josh Canham.
The Reds have notched impressive wins over the Highlanders and the previously unbeaten Brumbies in the last two weeks.
Flyhalf Carter Gordon missed the side's opening-round loss to the Waratahs with a leg complaint but has made an impact since, scoring the match-winner in Canberra.
Fellow flyhalf Tom Lynagh, as well as Test talents Hunter Paisami, Tate McDermott and Matt Faessler remain on the injured list.
'We've still got things to fix and work on for a big week against the Waratahs," Gordon said.
"I'm excited, the boys are excited, for this game. We've played them twice this year and we get another crack at them here."
Lock Angus Blyth will make his first Waratahs start against his former team while Jack Debreczeni will wear the No.10, ahead of another former Ballymore talent Lawson Creighton.
Triston Reilly will replace the injured Joseph-Aukuso Suaalii at outside centre while classy winger Sid Harvey will get his first start at Suncorp Stadium on Saturday.
